The IncuCyte S3 Live Cell Analysis System (Essence Biosciences) is a platform for analyzing living cell cultures at different time points. In this way one can obtain an automated quantification of the behavior of cells after disruption of selected genes and proteins. This IncuCyte has high-definition phase contrast lenses, and a software recognition package (ZOOM) that allows to mask cells, quantify and generate a time curve of cellular activities. As a result, cell activities, such as cell growth in 2D and 3D cultures, migration (wound healing and chemotaxis), cell aggregation (atmosphere formation), invasive clonogenicity, etc. can be observed and digitized. In addition, the cells can also be fluorescently labeled, whereby cell viability, apopotosis, phagocytosis and complex co-cultures can also be digitized. With this device both a single cell and complete cell populations can be studied. Important here is that this device is suitable for high throughput screening of different cellular processes in a 96- and 384-well format. The big advantage of this technology is that we can obtain kinetic information, which is impossible with the classical endpoint testing.
