Project description

One reason for drug failure at clinical stage is the lack of data supporting the drug target. We established a target screening and characterization platform, CRISParray, for arrayed high-throughput target perturbation with CRISPR technology, followed by RNA-sequencing. Owing to our platform’s arrayed configuration, we are in a unique position to integrate additional readouts. Studies have demonstrated the complementary between high content imaging and transcriptome profiling to uncover the function of a specific target. Here, we aim to integrate high content cell imaging through the CellPaint technology as an additional readout of the CRISPRarray platform. We will generate proof-of-concept data demonstrating the compatibility of CellPaint with the current implementation of CRISPRarray and optimize experiment setup and reaction conditions to tune workflow efficiency. We will develop software to combine RNA-seq and CellPaint readouts and further support target characterization.
