Project description

The various groups need appropriate technology to characterize, monitor and isolate specific gene-targeted and/or manipulated PSCs - down to the single-cell level for clonal selection - in addition to their specific differentiated progeny within a mixture of contaminating cells.

Therefore, we request a high-speed BD FACSAria Fusion cell sorter that permits analysis and sorting based on more than 16 different parameters. Up to 4 populations can be sorted in bulk and by incorporating an Automated Cell Deposition Unit (ACDU), direct and single-cell sorting into multiwell plates is feasible. Since the FACS machine is integrated with a biosafety cabinet, accidental exposure to users and/or samples is prevented. To analyze, follow and document our PSCs, two EVOS (fluorescence) cell imaging systems are also required since this allows evaluation of reporter gene expression without manipulation and disruption of the stem cell cultures.
