Project description

The candidate will focus on the research mandate of developing an efficient transformation and expression system for genetic manipulation of Candida bombicola. Further, it will produce the research to focus on the improvement of the cytochrome P450 monooxygenase activity, with the ultimate aim of an increased production amount, and other types of sophorolipids. For the production of more specific glycolipids challenging, multiple genetic modifications are required in a single strain. Given the limited availability of genetic markers for this purpose will be sought additional markers. One of the objectives for the creation of new glycolipids is the modification of the sophorolipide. The length of natural sophorolipids has 16 to 18 carbon atoms, and is determined by the specificity of the cytochrome P450. Candida bombicola P450's are supported by NADPH cytochrome P450 oxidoreductases, which demonstrated that the electron transport may form a rate limiting step. Therefore examined whether overexpression of CPR's may waive this limiting aspect and therefore can increase production sophorolipids.
